In the steady state most of the reactions involved in drug metabolism obey MichaelisMenten kinetics, at least approximately: Reaction rate = 7 mmaa x c ^ (3) where C is the concentration of the drug at the reaction site, and VmSiX and Km are constants of the reaction. Dedrick et al. (1972) have shown that when these constants are determined in vitro they may be used successfully to fit models to in vivo pharmacokinetic data. The chief processes affecting the transport of a drug from its portal of entry into the body to its receptors are described in the following sections.
